Parthenogenetic Activation of Porcine Oocytes by Calcium Ionophore A23187

چکیده
This study was designated to clarify the influence of activation of porcine matured oocytes by calcium ionophore on in vitro development of the parthenotes. The follicular oocytes were matured, activated and cultured in North Carolina State University-23 (NCSU-23) medium supplemented with 10% porcine follicular fluid (pFF). The in vitro-matured oocytes were exposed to calcium ionophore at concentrations of 12.5, 25 or 50 μM for 3, 5, 7 or 9 min. The activation rate of the oocytes increased as concentration of ionophore decreased, being at 27-33 and 6877 % for the oocytes treated with 50 and 12.5 μM ionophore, respectively. Almost all activated oocytes were haploid. The highest cleavage rate (76%) and developmental rate to morula (41%) were observed in the oocytes treated with 12.5 μM ionophore for 5 min. However, development to blastocyst was observed only in the oocytes treated with 25 μM ionophore for 3 and 5 min (3 and 4% of treated oocytes, respectively). We concluded that the activation treatment of the porcine oocytes with 12.5 μM ionophore for 5 min provided the highest developmental rate to morula, but this treatment is not sufficient to overcome a developmental block at the morula stage.

P-94: The Effect of Calcium Ionophore A23187 and Ethanol on Parthenogenetic Activation of Mouse Oocytes in Presence of Hydrostatic Pressure and Cy-tochalasin B
Background: Parthenogenetic activation of mammalian oocytes using artificial stimuli is commonly used in various reproductive bio-techniques. Calcium ionophore is known to elevate intracellular calcium levels in the cytoplasm of oocytes through the influx of calcium from extracellular spaces. Ethanol promotes a single intracellular Ca2+ increase of greater and longer amplitude than the initial ...

Background: Parthenogenetic activation is a possible way to produce homogeneous embryos with the some ploidy. Probably such embryos could be used in other areas of biotechnology. Calcium signals are known as important regulators of oocyte activation. Extracellular calcium is required for initiation of meiotic resumption and development. Calcium ionophore A23187 is known to elevate intracellular...
متن کاملP-76: Cytogenetic Investigation of Parthenogenetic Mouse Embryos Generated from In Vitro Activated Oocytes by Hydrostatic Pressure in The Presence of Calcium Ionophore and Ethanol
Background: The advances in cytogenetic techniques during the last few years have permitted not only the study of large populations of wild and domestic animals, but also the detection of chromosome anomalies in embryos. Chromosomal abnormalities are the most common cause of embryonic and fetal mortality in mammals. Most reports of chromosome anomalies in parthenogenetic embryos describe numeri...
متن کاملEffects of Thioglycolic Acid on Parthenogenetic Activation of Xenopus Oocytes
BACKGROUND Existing in Permanent-wave solutions (PWS), thioglycolic acid (TGA) is widely used in hairdressing industry for its contribution to hair styling. However, the toxicity of TGA, especially its reproductive toxicity, gradually calls the attention of more and more researchers. METHOD In this work, xenopus oocytes were pretreated with different concentration of TGA, and then activated b...

The objective of this study was to examine the effect of different sera and serum-like substances on the preimplantation development of porcine parthenogenetic embryos. Chemically activated (calcium ionophore A23187+cytochalasin B) pig oocytes were pre-cultured for five days. On day 5, the parthenogenetic embryos were treated with porcine follicular fluid (PFF), fetal bovine serum (FBS), horse ...
